01—“Pharmacopoeia of the People’s Republic of China (2020 Edition)”【Chinese Name】Poria【Pinyin】Fuling【English Name】PORIA【Introduction】This product is the dried sclerotium of the fungus Poria cocos (Schw) Wolf, belonging to the family Polyporaceae. It is harvested mainly from July to September. After excavation, the soil and sand are removed, and it is piled up to “sweat” before being spread out to dry until the surface is dry, then “sweated” again, repeatedly until wrinkles appear and most of the internal moisture is lost, then it is dried in the shade, referred to as “Fuling ge”; or fresh Poria is cut according to different parts and dried in the shade, referred to as “Fuling kuai” and “Fuling pian” respectively.【Characteristics】Fuling ge is spherical, oval, flat, or irregular clumps, varying in size. The outer skin is thin and rough, brown to dark brown, with obvious wrinkled texture. It is heavy, solid, with a granular fracture surface, some with cracks, the outer layer is light brown, the inside is white, and a few are light red, some contain pine roots. It has a faint odor, a bland taste, and is sticky when chewed.Fuling kuai is cut from peeled Poria, in cubic or thick slice form, varying in size. It is white, light red, or light brown.Fuling pian is cut from peeled Poria, in irregular thick slices, varying in thickness. It is white, light red, or light brown.【Identification】(1) The powder of this product is grayish-white. Irregular granules and branched clumps are colorless and gradually dissolve in water and chloroform solution. The mycelium is colorless or light brown, slender, slightly curved, branched, with a diameter of 3–8μm, some up to 16μm.(2) Take a small amount of the powder, add one drop of potassium iodide iodine test solution, and it shows deep red.(3) Take 1g of the powder, add 50ml of ether, treat with ultrasound for 10 minutes, filter, evaporate the filtrate, and dissolve the residue in 1ml of methanol to prepare the test solution. Take 1g of Poria as a control herb and prepare the control herb solution in the same way. According to thin-layer chromatography (General Rule 0502), apply 2μl of each of the two solutions on the same silica gel G thin-layer plate, using toluene-ethyl acetate-formic acid (20:5:0.5) as the developing agent, develop, remove, dry, and spray with a mixed solution of 2% vanillin-sulfuric acid solution-ethanol (4:1), heating at 105°C until the spots are clearly colored. Rats given 150, 300, and 600mg/kg of Poria Pi ethanol extract by gavage can increase urine volume by 29%, 21%, and 42%, respectively; K+excretion decreased by 18%, 22%, and 27%, respectively; in the 600mg/kg group, urine Na+excretion increased by 14%. Immune-Regulating EffectPoria water extract12g/kggiven to mice by gavage for 21 days can increase the number of peripheral T lymphocytes α-ANAE positive lymphocytes, enhance the proliferation response of splenic lymphocytes to ConA stimulation, increase the activity of IL-2 in the spleen of mice, and the activity of superoxide dismutase (SOD) in the liver of mice, inhibiting the generation of malondialdehyde (MDA), showing significant free radical scavenging effects. Poria water extract by gavage can significantly inhibit the 2,4-dinitrofluorobenzene-induced allergic contact dermatitis in mice, reducing ear swelling and ear tissue block weight, showing a certain dose-effect relationship. Mice given Poria polysaccharides200mg/kgby gavage for two weeks show significant improvement in cyclophosphamide-induced immune deficiency, increasing the proportion of CD3+cells in the Peyer’s patches, mesenteric lymph nodes, and spleen, and decreasing the proportion of CD19+cells, indicating that Poria polysaccharides have a regulatory effect on intestinal mucosal immunity.[3]In BALB/c mice immunized with ovalbumin5mg/kgby gavage for 1–3 weeks, starting one week before immunization with Poria polysaccharides200mg/kg, can significantly increase the secretion of ovalbumin-specific secretory immunoglobulin A (sIgA) in feces, and significantly increase the expression of co-stimulatory molecules CD80 and CD86 in the Peyer’s patches of the small intestine, indicating that Poria polysaccharides have a regulatory effect on intestinal sIgA secretion, related to its activation of Peyer’s patch B lymphocytes.[4]3. Anti-Transplant Rejection EffectPoria alcohol extract shows significant inhibition of acute rejection reaction in heart transplantation, with rats receiving Poria alcohol extract25mg/kgor50mg/kgby gavage showing significantly prolonged survival time of transplanted hearts, reduced pathological damage, and decreased levels of peripheral blood IL-2 and IFN-γ, as well as the percentages of CD3+, CD4+, and CD8+cells, and the CD4+/CD8+ratio decreased. Poria polysaccharides50mg/kggiven by gavage can significantly prolong the survival time of kidney transplant recipients in rats, significantly reduce pathological damage; peripheral blood IL-2, IFN-γ levels, and the percentages of CD4+and CD8+cells significantly decreased, indicating that Poria polysaccharides have a good inhibitory effect on acute rejection reactions in kidney transplantation.[5]4. Anti-Tumor EffectPoria contains various components with anti-tumor activity, including Poria polysaccharides, which not only have direct anti-tumor effects but also enhance the body’s immune function to exert anti-tumor effects. Poria polysaccharides50mg/kggiven every other day by intraperitoneal injection for 4 times can induce TNF, thus having a certain killing effect on tumor cells. Poria polysaccharides100mg/kgor150mg/kggiven by gavage to mice can significantly inhibit the growth of S180 and Ehrlich ascites carcinoma (EAC) tumors, reduce tumor weight, and significantly enhance the activity of NK cells in tumor-bearing mice. Derivatives of Poria polysaccharides also show good anti-tumor effects. Carboxymethyl Poria polysaccharides(CMP)25–500mg/kgintraperitoneally injected show an inhibition rate of75.5%–92.7%; CMP5–50mg/kgintravenously injected shows an inhibition rate of34.8%–61.0%; CMP25–100mg/kgintravenously injected shows an inhibition rate of20.1%–36.7%against H22 liver cancer in mice. Sulfated Poria polysaccharides show both killing and apoptosis-inducing effects on S180 tumor cells, with an inhibition rate of38.39%, and enhance the body’s immunity. Sulfated new Poria polysaccharides, while not enhancing the phagocytic function of mouse mononuclear macrophages, show significant anti-tumor effects. Poria polysaccharides also show anti-tumor metastasis effects, with Lewis lung cancer mice showing a significant reduction in the number of lung metastases after gavage of Poria polysaccharides(0.5mg/mouse).【5】5. Anti-Aging and Anti-Fatigue EffectsPoria water extract can increase the content of hydroxyproline in the skin. Elderly rats given Poria water extract2.4and8g/kgby gavage show significantly increased skin hydroxyproline content. Elderly rats given Poria polysaccharides12mg/kgby gavage for 10 consecutive days show significantly increased serum superoxide dismutase (T-SOD, CuSOD) activity and decreased malondialdehyde (MDA) content. Normal mice given Poria polysaccharides20and40mg/kgby gavage three times every 12 hours show significantly prolonged survival time in low-temperature swimming tests.6. Sedative, Hypnotic, and Memory Improvement EffectsPoria water decoction5.85g/kggiven by gavage for 7 consecutive days significantly increases the sleep rate of animals at subthreshold doses of pentobarbital sodium and prolongs the sleep time of mice at threshold doses of pentobarbital sodium.[7]; 42.8g/kggiven by gavage can significantly shorten the latency to reach the platform in the water maze experiment and reduce the activity of acetylcholinesterase in the brains of mice with memory impairment induced by scopolamine.[8]【7】7. Lipid-Lowering and Liver-Protective EffectsRats fed a high-fat diet and given Poria water decoction15gper kgdaily by gavage for 10 weeks, combined with aerobic exercise training, can significantly inhibit lipid abnormalities, reduce the degree of endothelial and vascular wall damage, and delay the development of atherosclerosis in rats.[9,10]Carboxymethyl Poria polysaccharides show antiviral effects on Hepatitis B virus-infected HepG2.2.15 cell lines, with half-maximal effective concentrations (IC)50of4.38and5.78mg/mlfor inhibiting HBsAg and HBeAg secretion, respectively. Using a carbon tetrachloride combined factor method to create a rat liver fibrosis model, gavage of carboxymethyl Poria polysaccharides40mg/ratonce daily for 2 weeks can significantly reduce the expression of TGF-β, caspase-3, type II collagen, and laminin in liver tissues; reduce the expression of Smad-3 in liver fibrosis rats and increase the expression of Smad-7, indicating significant anti-liver fibrosis effects.[11]Using carbon tetrachloride, high-fat low-protein diet, and alcohol to replicate a rat liver cirrhosis model, subcutaneous injection of Poria alcohol extract75mg/kgonce daily for 3 weeks can significantly reduce the degree of liver fibrosis and lower collagen content in the liver.8. Other Pharmacological Effects100% Poria extract filter paper shows inhibitory effects on Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, anthrax bacillus, Escherichia coli, group A streptococcus, and group B streptococcus. Poria water extract can induce an increase in intracellular calcium ion concentration, with concentrations of Poria water extract at31–250mg/Linducing an increase in intracellular calcium ion concentration of9.9%–33.7%, increasing with the dosage. Poria has important effects on the function of mitochondria and microtubule structure in nerve cells, with Poria water extract10–20mg/Lincubated with cells for24hours significantly resisting the reduction of mitochondrial MTT reduction ability caused by sodium azide.9. Toxicological StudiesThe median lethal dose (LD50) of carboxymethyl Poria polysaccharides injected intravenously in mice is3.13g/kg±0.14g/kg, with symptoms of reduced activity, limb weakness, convulsions, and weak breathing before death, occurring within1–2days. Mice given carboxymethyl Poria polysaccharides15g/kgonce by gavage show no toxic reactions. Intravenous injection of carboxymethyl Poria polysaccharides157and313mg/kgonce daily for 3 months shows no significant toxic reactions. Pregnant rats injected intraperitoneally with carboxymethyl Poria polysaccharides from day 6 to 15 of pregnancy show no fetal malformations. The LD50of sulfated Poria polysaccharides is7.358g/kg, with a 95% confidence interval of7.358g/kg±0.894g/kg.Appendix:1. Poria Pi This product is the dried outer skin of Poria.
